The enzyme substrate method uses coliform producing beta galactosidase to decompose the color source substrate ONPG (Ortho-nitrophenyl-β-D-galactopyranoside) to make the culture medium change yellow,Escherichia coli produces β-glucuronidase to decompose the fluorescent chromatic source substrate MUG( 4-methyl-umbelliferyl-β-glucuronide)produce fluorescence under 366 nm ultraviolet, The principle is used to determine the quality and quantity of total coliform bacteria, fecal coliform (heat coliform bacteria) and Escherichia coli in water. When a few non target bacteria|in the water sample try to react with ONPG or MUG to interfere with the detection results, the matrix of the special formula in the colitech medium can inhibit the reaction process and control the false positive. At the same time, these special formulations can effectively promote the reaction of the biological enzymes and the fixed substrates of the target bacteria. The incidence of false negative is lower than that of the traditional method. Colitech enzyme substrate test reagent is suitable forGB/T5750.12-2006"drinking water standard detection method"medium. The combined use of Colitech enzyme substrate reagent and 51 or 97 detection plates can be used to detect target bacteria in 100 ml water samples, the detection range of 51 detection plates is 0~200MPN,and the detection range of 97determination disk is 0~ 2419MPN,without dilution.
97 Well Detection Plate
The 97 well detection plate is used with the colitech enzyme substrate method to determine the MPN value of the coliform bacteria in 100ml water accurately. The reagent and water sample were dissolved in the hole detection plate after dissolving the reagent and water sample according to the colitech enzyme substrate method, and the positive pore lattice was counted with the program controlled quantitative sealer. The value of MPN concentration in the water sample was calculated by 97 wells MPN table.
97 Well Detection Plate Operation description
1.A single 97 well detection plate is used to make the hole facing the palm
2.Press the upper part of the hole detection plate by hand to make the inspection orifice bend to the palm.
3.Pull the aluminum foil and pull the aluminum foil to separate the holes.Avoid contact with the inside of the quantitative
detection plate by hand.
4.The reagent and water sample are dissolved and then poured into the quantitative detection tray. Avoid contacting the aluminum
foil with the solution and pat the orifice to remove bubbles.
5.The 97 well test of reagent and water sample has been poured down.After fitting with the 97 well rubber liner, the opening is
pushed upward into the sealing of the program controlled quantitative sealer.

8.count the number of positive holes in big holes and small holes, and check the 97 well MPN table count.
